BACKGROUND: Early postnatal overfeeding (PO) induces long-term overweight and reduces brown adipose tissue (BAT) thermogenesis. Exercise has been suggested as a possible intervention to increase BAT function. In this study, we investigated chronical effects of moderate-intensity exercise in BAT function in postnatal overfed male Wistar rats METHODS: Litters' delivery was on postnatal-day 0 - PN0. At PN2, litters were adjusted to nine (normal litter - NL) or three pups (small litter - SL) per dam. Animals were weaned on PN21 and in PN30 randomly divided into sedentary (NL-Sed and SL-Sed) or exercised (NL-Exe and SL-Exe), N of 14 litters per group. Exercise protocol started (PN30) with an effort test; training sessions were performed three times weekly at 60% of the VO2max achieved in effort test, until PN80. On PN81, a temperature transponder was implanted beneath the interscapular BAT, whose temperature was assessed in periods of lights-on and -off from PN87 to PN90. Sympathetic nerve activation of BAT was registered at PN90. Animals were euthanized at PN91 and tissues collected RESULTS: PO impaired BAT thermogenesis in lights-on (pPO < 0.0001) and -off (pPO < 0.01). Exercise increased BAT temperature in lights-on (pExe < 0.0001). In NL-Exe, increased BAT activity was associated with higher sympathetic activity (pExe < 0.05), ß3-AR (pExe < 0.001), and UCP1 (pExe < 0.001) content. In SL-Exe, increasing BAT thermogenesis is driven by a combination of tissue morphology remodeling (pExe < 0.0001) with greater effect in increasing UCP1 (pExe < 0.001) and increased ß3-AR (pExe < 0.001) content. CONCLUSION: Moderate exercise chronically increased BAT thermogenesis in both, NL and SL groups. In NL-Exe by increasing Sympathetic activity, and in SL-Exe by a combination of increased ß3-AR and UCP1 content with morphologic remodeling of BAT. Chronically increasing BAT thermogenesis in obese subjects may lead to higher overall energy expenditure, favoring the reduction of obesity and related comorbidities.

The consumption of fructose has increased in children and adolescents and is partially responsible for the high incidence of metabolic diseases. The lifestyle during postnatal development can result in altered metabolic programming, thereby impairing the reproductive system and fertility during adulthood. Therefore, the aim of this study was to evaluate the effect of a high-fructose diet in the male reproductive system of pubertal and adult rats. Male Wistar rats (30 d old) were assigned to four different groups: Fr30, which received fructose (20%) in water for 30 d and were euthanized at postnatal day (PND) 60; Re-Fr30, which received fructose (20%) for 30 d and were euthanized at PND 120; and two control groups C30 and Re-C30, which received water ad libitum and were euthanized at PND 60 and 120, respectively. Fructose induced an increase in abnormal seminiferous tubules with epithelial vacuoles, degeneration, and immature cells in the lumen. Moreover, Fr30 rats showed altered spermatogenesis and daily sperm production (DSP), as well as increased serum testosterone concentrations. After discontinuing high-fructose consumption, DSP and sperm number decreased significantly. We observed tissue remodeling in the epididymis, with a reduction in stromal and epithelial compartments that might have influenced sperm motility. Therefore, we concluded that fructose intake in peripubertal rats led to changes in the reproductive system observed both during puberty and adulthood.

Social life necessitates cognitive competence to meet the dynamic demands of social development. The formation of dominance hierarchy is a general phenomenon in social groups. As an essential element of executive and cognitive function, working memory could influence and be influenced by social status in a dominance hierarchy. However, the direction and degree of the association between them through different developmental stages remain unclear. To address this issue and clarify the "cause or consequence" problem, we investigated the spatial working memory performance in a Y-maze and Morris water maze in home-caged sibling Wistar rats (N = 26 cages, three rats/cage) through three stages of their life: before (week 7), during (week 10), and after (week 20) assumed timings of the social dominance hierarchy formation (SDHF). We used the social dominance tube test during the assumed time of hierarchy formation (weeks 9-11) to measure the relative dominance status in each cage. Here, we found that higher working memory index before SDHF could be predictive of later acquisition of higher social status. Working memory performance declined for all animals during SDHF, in which agonistic conflicts are increased. However, living within an established hierarchical social network for several weeks deteriorated the working memory performance of dominant and middle-ranked animals, while the performance of subordinates improved and got significantly better than higher-ranked animals. In conclusion, while working memory and social status were correlated positively before dominance hierarchy formation, there was a trade-off between them after the formation of it. In contrast to the common view, these results highlight the adverse effect of higher social status on cognitive behavior.

Tetrabromobisphenol A (TBBPA) has become a ubiquitous indoor contaminant due to its widespread use as an additive flame retardant in consumer products. Reported evidence of endocrine disruption and accumulation of TBBPA in brain tissue has raised concerns regarding its potential effects on neurodevelopment and behavior. The goal of the present study was to examine the impact of developmental TBBPA exposure, across a wide range of doses, on sexually dimorphic non-reproductive behaviors in male and female Wistar rats. We first ran a pilot study using a single TBBPA dose hypothesized to produce behavioral effects. Wistar rat dams were orally exposed using cookie treats to 0 or 0.1 mg TBBPA/kg bw daily from gestational day (GD) 9 to postnatal day (PND) 21 to assess offspring (both sexes) activity and anxiety-related behaviors. Significant effects were evident in females, with exposure increasing activity levels. Thus, this dose was used as the lowest TBBPA dose in a subsequent, larger study conducted as part of a comprehensive assessment of TBBPA toxicity. Animals were exposed to 0, 0.1, 25, or 250 mg TBBPA/kg bw daily by oral gavage starting on GD 6 through PND 90 (dosed dams GD 6 - PND 21, dosed offspring PND 22 - PND 90). Significant behavioral findings were observed for male offspring, with increased anxiety-like behavior as the primary phenotype. These findings demonstrate that exposure to environmental contaminants, like TBBPA, can have sex-specific effects on behavior highlighting the vulnerability of the developing brain.

During the lactation period, rat pups are fed by the dam, and the patterns of mother-pup interaction change during this period. Additionally, there are changes in feeding; first, mother´s milk is the only food needed for sustenance, and later, it is combined with solid food and water. GH serum concentrations depend on both maternal-pup interaction and energy metabolism. In the artificial rearing (AR) procedure, pups are deprived of mother-pup interaction, and the feeding pattern is controlled. This rearing paradigm has been used in rats to analyze the effects of maternal deprivation on social behavior. In the present study, we analyzed the variation in GH, acylated ghrelin and IGF-1 serum concentrations throughout the lactation period in AR pups. At pnd7, the maternal rearing (MR) pups responded to a 4 h fast with a drop in GH serum concentration, which is a well-known response to maternal deprivation. GH serum levels in the AR pups did not change, suggesting an adaptation phenomenon. A dopamine inhibitory effect of GH secretion was observed in pnd7 cultured somatotropes, suggesting dopamine regulation of GH secretion at this age. Acylated ghrelin serum levels in the AR pups showed an inverted pattern compared to that in the MR pups, which was related to the artificial feeding pattern. IGF-1 serum levels were lower in the AR pups than in MR pups, which was associated with hepatic GH resistance and with low Igf1 mRNA expression at pnd7. Interestingly, at pnd14, both pup groups showed high hepatic Igf1 mRNA expression but low IGF-1 serum levels, and this was inverted at pnd21. However, serum glucose levels were lower in the AR pups at pnd14 but reached the same levels as the MR pups at pnd21. Moreover, hepatomegaly and higher hepatic GH-receptor levels were observed in the AR pups at pnd21, which was in agreement with an absence of a solid food meal. During AR, the pups lost the maternal interaction-stimulated GH secretion, which correlated with lower IGF-1 serum levels during the first week of postnatal development. Later, the AR pups exhibited hepatic responses, in order to satisfy the metabolic demand for the normal weaning, with low carbohydrates levels in their meal.

Se realizó un estudio descriptivo, observacional y transversal en 160 cortes histológicos de la fascia dentada del hipocampo de ratones BALB/c y ratas Wistar blancas, en el Laboratorio de Investigaciones Biomédicas de la Universidad de Ciencias Médicas de Santiago de Cuba, de septiembre de 2013 a igual mes de 2014, con vistas a determinar las transformaciones histológicas que ocurren en dicha fascia en el segundo y sexto días posnatales. La observación microscópica de estos cortes se realizó empleando del software Image J. La extensión de la fascia al sexto día de vida llegó a ser mayor en los ratones; los máximos incrementos del grosor en ambos tipos de roedores se encontraron en el hilus, y el estrato granuloso fue de menor crecimiento en las ratas. La celularidad en los roedores presentó mayores proporciones en las tres regiones del hilus al segundo día, pero disminuyó en el sexto día, mientras que las zonas relacionadas con el hilus mantuvieron una mayor cantidad de células; sin embargo, el número celular disminuyó en ambas regiones moleculares de la fascia de las ratas.


A descriptive, observational and cross-sectional study was carried out in 160 histological cuts of the hippocampus fascia dentata from mice BALB/c and rats white Wistar, in the Laboratory of Biomedical Investigations from Santiago de Cuba Medical University, from September, 2013 to the same month in 2014, with the aim of determining the histological transformations that take place in this fascia in the second and sixth posnatal days. The microscopic observation of these cuts was carried out using the software Image J. The extension of the fascia at the sixth day of life was larger in the mice; the maximum increases of thickness in both types of rodents were in the hilus, and the granular stratum was of smaller growth in rats. The celularity in the rodents presented larger proportions in the three regions from the hilus at the second day, but it decreased at the sixth day, while the areas related to the hilus maintained a greater quantity of cells; however, the cellular number diminished in both molecular regions of the rats fascia.

Renin angiotensin aldosterone system (RAAS) plays an essential role in the homeostatic control of arterial blood pressure, perfusion of tissues, and control of extracellular fluid. Its components are highly expressed in the developing kidney, general vasculature, brain, and heart. A modified intrauterine environment alters mechanisms controlling blood pressure (BP) and can lead to hypertension in the adult offspring and developmentally programmed RAAS can be involved in this process. There are very little data about the effects of increased angiotensin II (Ang II) concentrations during pregnancy on in utero development of the fetus. In our study, we administered Ang II to pregnant female rats via osmotic mini-pumps and evaluated the postnatal development and BP control in the offspring. To estimate possible developmental changes in sensitivity to salt, we exposed the offspring to a diet with increased salt content and measured plasma aldosterone levels and plasma renin activity. Increased Ang II during pregnancy raised BP in the offspring; however, salt sensitivity was decreased in comparison to controls. Relative weight of the left ventricle was decreased in the offspring prenatally exposed to Ang II, while relative kidney weight was reduced only in female offspring. Prenatal treatment led to increased aldosterone levels and decreased plasma renin activity, suggesting a complex physiological response. Our results suggest that conditions leading to upregulation of RAAS during pregnancy can influence the cardiovascular system of the fetus and have a long-term impact on the offspring's health.

A atividade elétrica cardíaca pode ser registrada por meio do eletrocardiograma (ECG), registro capaz de captar as alterações do percurso elétrico. Na literatura científica, não existe descrição de padrões eletrocardiográficos de ratos Wistar desde a fase de recém-nascidos até serem considerados envelhecidos. O objetivo do presente estudo foi avaliar o padrão eletrocardiográfico de ratos Wistar desde a fase de recém-nascidos até serem considerados envelhecidos, no intuito de que os resultados possam contribuir como referencial dos padrões da espécie. Foram utilizados ratos Rattus novergicus albinus, da linhagem Wistar. Os ratos foram divididos em cinco grupos experimentais, sendo definidos de acordo com a idade e assim denominados: grupo 21 dias (21D), dois meses (2M), três meses (3M), sete meses (7M) e 18 meses (18M), sendo n=20 por grupo. Para a realização do ECG, eletrodos conectados aos canais do eletrocardiógrafo foram posicionados nos animais e captaram o sinal elétrico do coração em seis derivações. Os dados foram agrupados e submetidos a comparação por meio de ANOVA e teste de Tukey-Kramer, P≤0,05. Os resultados mostraram que, de forma geral, as modulações ao longo do processo de envelhecimento diante dos parâmetros relacionados ao ECG se iniciam a partir do período 2M, com aparente estabilidade até 3M e/ou 7M, seguindo por exacerbação das modulações em 7M e/ou 18M, a depender da variável eletrocardiográfica em questão. Em relação à frequência cardíaca (FC), concomitantemente, nota-se um padrão similar, ou seja, diminuição dela a partir do período 2M, com estabilidade até 7M, seguindo por diminuição mais pronunciada em 18M. Os resultados sugerem que, devido ao processo de envelhecimento, o coração de ratos Wistar apresenta modificações na condução do estímulo elétrico, bem como redução na FC, fatores que merecem atenção enquanto indicadores de inclusão/exclusão de animais em grupos experimentais por parte de pesquisadores que utilizam a referida espécie como modelo de estudo.(AU)


The cardiac electrical activity can be recorded through the electrocardiogram (ECG), this recording is able to capture the changes in the electric route. In the scientific literature, there is no description of electrocardiographic patterns of Wistar rats at the stage of newborn to be considered aged. The aim of this study was to evaluate the electrocardiographic pattern of Wistar rats at various stages of growth, in order for the results to contribute as a standard reference. Rattus norvegicus albinus rats of the Wistar lineage were used. The rats were divided into 5 experimental groups being defined according to age and named as follows: group 21 days (21D), 2 months (2M), 3 months (3M), 7 months (7M) and 18 months (18M), where n = 20 per group. For the realization of ECG, electrodes connected to the electrocardiograph channels were positioned in animals to capture the electrical signal of the heart in 6-lead. The data were grouped and subjected to comparison by ANOVA and Tukey-Kramer test, P=0.05. The results showed that, overall, the modulations along the front aging process, related to the ECG, are initiated from 2M period, apparent stability by 3M and/or 7M, followed by exacerbation of modulations in 7M and/or 18M, depending on the electrocardiographic variable in question. Front of heart rate (HR), concomitantly, there is a similar pattern, that is, decreasing from 2M period with stability until 7M, followed by more pronounced decrease in 18M. The results suggest that due to the aging process, the heart of Wistar rats shows changes in the conduction of electrical stimulation and reduction in heart rate, factors that deserve attention as indicators of inclusion / exclusion of animals in experimental groups by researchers using that species as a model.(AU)

The nutraceutical industry has proliferated in recent years, with the most popular form of supplementation being the multivitamin-multimineral (MVMM) supplement. In the animal health sector, supplement use has also expanded. The objective of this study was to determine the effects of MVMM supplementation, beneficial or otherwise, on the general health status of the spontaneously hypertensive rat (SHR) strain, an animal model used in hypertension research. A commercially prepared MVMM supplement was given tri-weekly via oral dosing for 8 weeks to two groups of seven adult female SHR and Wistar rats. Their corresponding control groups were dosed with deionised water only. Systolic and diastolic blood pressure, fasting blood glucose, growth rate and food and water intake were measured weekly. At the end of 8 weeks, the animals were euthanased and a full blood profile, urine sodium to potassium ratio, blood urea nitrogen levels and total plasma cholesterol was measured for all groups. The results indicated that growth rate was higher for the SHR supplemented group. Supplementation also decreased diastolic blood pressure in both Wistar and SHR groups and increased red blood cell count and decreased total cholesterol in the SHR group. No adverse effects on the general health status of the animals were observed. MVMM supplementation may therefore be useful in aiding growth and delaying the onset of hypertension and its effects. It may also assist in the longevity of the breeding stock of SHR rats.

Female Wistar rats are frequently used in experimental models to study hormone and bone pathologies and treatments. Most experimental studies involving histomorphometric evaluation assessed long bones, and few reports also studied mandibular bone. The aim of this work was to clarify and distinguish the age-related histomorphometric changes that occur in the tibia (subchondral bone) and in the mandible (interradicular bone), and thus obtain reference histomorphometric data of healthy female Wistar rats at different growth stages. Three groups of 8 healthy female Wistar rats were euthanized at 6 (GI), 10 (GII), and 14 (GIII) weeks. The tibiae and mandible were resected and histologically processed to obtain H&E stained sections of the tibia and the lower first molar to analyze the following histomorphometric parameters: Bone volume, trabecular width, trabecular number (Th.N)(1/mm), growth cartilage width, hypertrophic cartilage width and number of osteoclasts per area in the tibiae, and bone volume and number of osteoclasts per area N.Oc/mm(2) in the interradicular bone of the first lower molar. A significant decrease in subchondral bone volume as a result of a decrease in trabecular number and growth cartilage width was observed in 14-week-old rats. Conversely, interradicular bone volume was found to increase with age. The results highlight the importance of analyzing both types of bone to better understand the response of two different trabecular bones, contributing in turn to decision making regarding treatment strategies and disease management.

Articular cartilage (AC) covers the surface of bones in joints and functions as a cushion against mechanical loading. The tissue contains abundant extracellular matrix (ECM), which mainly consists of proteoglycans (PG) and collagen (COL) fibres. The property of AC is gradually changing by ageing with gravity loading. To know the property change of AC by initial gravity loading during short period after birth, we performed histological assays and proteomics assay on the AC of the femoral condyle in knee joints of perinatal rats. The water content (%) was significantly decreased in neonate AC compared with fetal AC. During the perinatal stages (E19 and P0), the localizations of glycosaminoglycan (GAG) and type I and II COLs were homogeneous. The density of chondrocytes was significantly decreased in the deeper layers comparing with the surface layer in neonate AC. In addition, we found a drastic change in the protein expression pattern on proteomic analysis. The expressions of ECM components were relatively increased in neonate AC compared with fetal AC.

PURPOSE: To investigate allometric relationships among body mass (BM), muzzle-tail length (MTL), and tibia length (TL) in Wistar rats and establish their growth rate change parameters. METHODS: Eighteen male and 18 female Wistar rats were studied from the 3rd to the 21st week of age. BM, MTL, and TL were measured daily, and relative growth was compared using allometry. RESULTS: A positive correlation between BM and MTL (p<0.05) and BM and TL (p<0.05) was observed. Males and females showed comparable curves; however, females had turning points at a younger age. The allometric relationship between BM and MTL presented a regular increase until reaching a mass of 351 g (males) and 405 g (females). BM and TL showed an initial increase until 185 g (males) and 182 g (females), and then reached a plateau that finished at 412 g (males) and 334 g (females), to display another increase. CONCLUSIONS: The allometric relationship of body mass with animal length and tibia length was comparable for male and female rats, with female rats maturing earlier. Animal longitudinal growth occurred in a single stage. In contrast, tibia length depicted two stages of accelerated growth with an intermediate period of deceleration.

Detecting neurodevelopµental disorders of cognition at the earliest possible stages could assist in understanding them mechanistically and ultimately in treating them. Finding early physiological predictors that could be visualized with functional neuroimaging would represent an important advance in this regard. We hypothesized that one potential source of physiological predictors is the spontaneous local network activity prominent during specific periods in development. To test this we used calcium imaging in brain slices and analyzed variations in the frequency and intensity of this early activity in one area, the entorhinal cortex (EC), in order to correlate early activity with level of cognitive function later in life. We focused on EC because of its known role in different types of cognitive processes and because it is an area where spontaneous activity is prominent during early postnatal development in rodent models of cortical development. Using rat strains (Long-Evans, Wistar, Sprague-Dawley and Brattleboro) known to differ in cognitive performance in adulthood we asked whether neonatal animals exhibit corresponding strain-related differences in EC spontaneous activity. Our results show significant differences in this activity between strains: compared to a high cognitive-performing strain, we consistently found an increase in frequency and decrease in intensity in neonates from three lower performing strains. Activity was most different in one strain considered a model of schizophrenia-like psychopathology. While we cannot necessarily infer a causal relationship between early activity and adult cognition our findings suggest that the pattern of spontaneous activity in development could be an early predictor of a developmental trajectory advancing toward sub-optimal cognitive performance in adulthood. Our results further suggest that the strength of dopaminergic signaling, by setting the balance between excitation and inhibition, is a potential underlying mechanism that could explain the observed differences in early spontaneous activity patterns.

Introducción: Las ratas Wistar son una de las cepas más populares y utilizadas cotidianamente para la investigación en el laboratorio sirviendo como una importante herramienta de investigación, por lo que, exige el control estricto de variables como la edad, el sexo y el peso corporal, y de esta forma poder extrapolar los resultados al modelo humano. Objetivo: Desarrollar curvas de referencia para valorar el crecimiento físico de ratas machos Wistar en función de la edad cronológica y la maduración somática desde una perspectiva no-invasiva. Metodología: Fueron estudiadas 731 ratas machos Wistar de forma transversal. Se evaluó la edad, peso corporal y la superficie corporal. Se utilizó el método LMS para construir curvas de percentil en función del peso y la maduración somática. Resultados: Las curvas de crecimiento físico propuestas sirven para realizar el seguimiento del crecimiento físico y el diagnóstico del estado nutricional de ratas machos de cepa Wistar. Los puntos de corte prepuestos son: P3, P10, P25, P50, P75, P90 y P97. Conclusión: Los resultados sugieren que los científicos de diversas áreas puedan usar tales referencias, con el objetivo de extrapolar las fases del crecimiento somático de la rata de laboratorio al modelo humano y es una alternativa no-invasiva para valorar el crecimiento y el estado nutricional (AU)


Introduction: Wistar rats are one of the most popular strains routinely used for research in the laboratory to serve as an important research tool, so it requires strict control of variables such as age, sex and body weight, and Thus to extrapolate the results to the human model. Objective: To develop reference curves for assessing the physical growth of male Wistar rats according to chronological age and somatic maturation from a non-invasive. Methodology: The subjects studied were 731 male Wistar rats transversely. We assessed age, body weight and body surface. LMS method was used to construct percentile curves based on weight and somatic maturation. Results: The proposed physical growth curves are used to track the physical growth and nutritional status diagnosis of male Wistar rats. Budgets by cutting points are: P3, P10, P25, P50, P75, P90 and P97. Conclusion: The results suggest that scientists from different areas can use such references, in order to extrapolate somatic growth phases of the laboratory rat and the human model is a non-invasive alternative to assess growth and nutritional status (AU)

INTRODUCTION: Biological age is an important parameter for growth and development assessment. It can be evaluated through the observation of radiographic changes in skeletal maturation of cervical vertebrae. OBJECTIVE: This study aims to: a) verify if there is correlation between growth curve and the stages of bone age of animals used in laboratories, by evaluating radiographs of the cervical vertebrae; b) correlate these stages with their correspondents in humans. METHODS: 35 Wistar rats were evaluated for a period of 160 days, starting at day 22nd (weaning), with cross sections for periodic weighing, length measurement and digital radiography. Radiographs of the cervical vertebrae (C2 and C3) were measured by means of a computer program (Radio IMP). Data were submitted to statistical analysis (ANOVA) and Pearson correlation. RESULTS: Growth spurt was characterized by fast increasing in weight and length. Through ANOVA, differences were observed in the cervical measurements between days 22, 97, 127, 157, 187 and 217 (p <0.001). A high correlation was found between increasing in body length and weight, as well as in cervical vertebrae height (r = 0.86). Increments in concavities of vertebrae were also observed, similar to humans. CONCLUSIONS: There is correlation between body growth and maturation of cervical vertebrae in rats. Despite the continuous development of concavities, it was not possible to clearly identify the 5/6 stages as in studies of cervical vertebrae maturation in humans.

The present study analyzes the interaction between prenatal stress and mother's behavior on brain, hormonal, and behavioral development of male offspring in rats. It extends to males our previous findings, in females, that maternal care can alter behavioral dimorphism that becomes evident in the neonates when they mature. Experiment 1 compares the maternal behavior of foster mothers toward cross-fostered pups versus mothers rearing their own litters. Experiment 2 ascertains the induced "maternal" behavior of the male pups, derived from Experiment 1 when they reached maturity. The most striking effect was that the males non-exposed to the stress as fetuses and raised by stressed foster mothers showed the highest levels of "maternal" behavior of all the groups (i.e., induction of maternal behavior and retrieving behavior), not differing from the control, unstressed, female groups. Furthermore, those males showed significantly fewer olfactory bulb mitral cells than the control males that were non-stressed as fetuses and raised by their own non-stressed mothers. They also presented the lowest levels of plasma testosterone of all the male groups. The present findings provide evidence that prenatal environmental stress can "demasculinize" the behavior, brain anatomy and hormone secretion in the male fetuses expressed when they reach maturity. Moreover, the nature of the maternal care received by neonates can affect the behavior and physiology that they express at maturity.

INTRODUÇÃO: a idade biológica é um parâmetro importante na avaliação do crescimento e desenvolvimento, podendo ser avaliada por meio da observação de alterações na maturação óssea das vértebras. OBJETIVO: o presente estudo visa descrever e relacionar a curva de crescimento de ratos utilizados em pesquisas laboratoriais com os estágios de idade óssea, avaliados por radiografias de vértebras cervicais, e correlacionar esses estágios com estudos correspondentes em humanos. MÉTODOS: foram avaliados 35 ratos Wistar em um período de 160 dias, iniciando no 22º dia de vida (desmame), com cortes transversais periódicos para pesagem, medição do comprimento e radiografias digitais. As radiografias das vértebras cervicais (C2 e C3) foram mensuradas por meio de um programa de computador (Radio IMP). Os dados foram submetidos à análise estatística de variância (ANOVA). RESULTADOS: o surto de crescimento caracterizou-se por aumento rápido de peso e comprimento, seguido por um período de crescimento lento e de estabilidade. Uma alta correlação (r = 0,86) foi verificada entre o aumento de peso e o comprimento do corpo, bem como o comprimento das vértebras cervicais. Incrementos nas concavidades das vértebras dos ratos foram observados, semelhantemente aos resultados obtidos em estudos em humanos. CONCLUSÕES: existe correlação entre o crescimento corporal de ratos e a maturação das vértebras cervicais. Apesar da detecção de desenvolvimento contínuo de concavidades das vértebras, não foi possível identificar claramente os 5 ou 6 estágios de maturação óssea descritos em seres humanos.

Fish and other aquatic organisms are important source of dietary proteins for the human population. Fish meat, however, is contaminated with methyl mercury (MeHg), a potent neurotoxin. The well known Minamata and Niigata epidemic outcomes in Japan have raised the awareness of the health risk resulting from consumption of fish (and shellfish) from water basins polluted with industrial wastes containing mercury. In the present study, pregnant rat dams were exposed to environmental toxic elements--methyl mercury, 1000-1200 h, daily from the fifth gestation day (GD5) till parturition. Three groups of animals were given, by gavages, MeHg (0.5, 1.0 and 2.0 mg/kg/day) and control group received 0.9% saline at the same time. All animals were allowed to deliver and wean their offspring. Pups were evaluated for early development effects. There was a significant effect of treatment on somatic growth such as reduction in percentage of maternal weight gain (20.62%) at higher dose level whereas there was no change in percentage of live birth (100.00%) with 0.5 and 1.0 mg/kg dose treatment groups. There was a significant increase in the percentage of resorption (100.00%) per litter with 2.0 mg/kg/day MeHg dose. Average gestation length (days) and percentage resorption per litter or percentage foetuses/malformations per litter were not affected at 0.5 and 1.0 mg/kg/day dose level. The results of the study confirmed the high-teratogenic potential of MeHg and the need of payng increased attention to MeHg concerning its exogenous use during pregnancy.

OBJECTIVE: The present study assessed the pregnancy and lactation performances of rats fed an experimental diet based on the foods listed in the Family Budget Survey (Pesquisa de Orçamento Familiar) 2002/2003 and the impact of said diet on the growth and development of the pups until weaning. METHODS: Wistar (n=12) rats were randomly divided into two groups: a control group (control group, n=6) fed a commercial chow (Labina®, Brazil) and an experimental group (n=6) fed the Family Budget Survey diet during the entire pregnancy and lactation period. All animals had free access to food and water during the entire study period. RESULTS: The Family Budget Survey diet increased the duration of pregnancy (control group=21.00±0.00; POFG=21.57±0.55, p=0.025) and made the dams lose weight during the lactation period (control group=27.92±18.47g; POFG=-15.66±16.90g). The Family Budget Survey group presented low food, energy and nutrient intakes during pregnancy, which became even lower during lactation. Pups from Family Budget Survey dams presented lower body weight at weaning (control group=52.38±4.49g; POFG=39.88±2.78g, p=0.001) and lower nose-to-anus length (control group= 117.37±0.64mm; POFG=125.62±0.96mm, p=0.001). However, some physical milestones and reflexes occurred earlier, such as the placing response reflex [control group= 12.00 (9.00-15.00) days; POFG=9.50 (9.00-14.00) days] aerial righting reflex [control group=18.00 (17.00-20.00) days; POFG=16.00 (13.00-18.00) days] and unfolding of the external ear [control group=3.00 (3.00-3.00) days; POFG=2.00 (2.00-3.00) days]. CONCLUSION: The Family Budget Survey diet seems to be more detrimental to the physical growth of the pups than to their brain growth, according to the assessed reflexes and physical milestones and measures. This may be due to the low protein content of the diet for rat reproduction and growth combined with adequate fat and essential fatty acid contents. Providing an adequate amount of protein to the Family Budget Survey diet may normalize pup growth.


OBJETIVO: O estudo se propõe avaliar os desempenhos de prenhez e lactação de ratas alimentadas com dieta experimental baseada nos alimentos adquiridos para consumo pela Pesquisa de Orçamento Familiar 2002-2003 e as repercussões no crescimento e desenvolvimento da prole até o desmame. MÉTODOS: Ratas Wistar (n=12) foram aleatoriamente divididas em dois grupos experimentais: um grupo-controle alimentado com dieta comercial Labina®, Brasil (n=6) ou dieta experimental (n=6) por toda gestação e lactação. Ração e água foram oferecidas ad libitum por todo período. RESULTADOS: A dieta Pesquisa de Orçamento Familiar na gestação e lactação aumentou a duração da gestação (grupo-controle=21,00±0,00; GPOF=21,57±0,55, p=0,025) e induziu uma variação negativa de peso nas mães ao final da lactação (grupo-controle=27,92±18,47g; POFG=-15,66±16,90g). Durante a gestação houve redução no consumo de alimentos, energia e nutrientes, exceto de lipídios no grupo experimental Pesquisa de Orçamento Familiar e esta foi intensificada na lactação. A prole proveniente das mães com dieta Pesquisa de Orçamento Familiar apresentou menor peso corporal ao desmame (grupo-controle=52,38±4,49g; POFG=39,88±2,78g, p=0,001) e menor comprimento naso-anal (grupo-controle=117,37±0,64mm; POFG=125,62±0,96mm, p=0,001). No entanto, o desenvolvimento físico e reflexo não mostrou diferenças em alguns resultados ou apresentou adiantamento como observado na colocação pelas vibrissas [grupo-controle: 12,00 (9,00-15,00) dias; POFG=9,50 (9,00-14,00) dias] e queda livre [grupo-controle:18,00 (17,00-20,00); POFG=16,00 (13,00-18,00) dias], bem como da abertura do pavilhão do conduto auditivo [grupo-controle=3,00 (3,003,00); POFG=2,00 (2,00-3,00) dias]. CONCLUSÃO: A dieta Pesquisa de Orçamento Familiar parece causar maior agressão ao crescimento somático que ao desenvolvimento cerebral da prole avaliado pela ontogênese reflexa e os caracteres físicos. Estas resultantes podem estar associadas a uma possível inadequação proteica da dieta para o período de reprodução e crescimento dos ratos em conjunto com suficiente aporte de lipídeos e ácidos graxos essenciais. Sugere-se que uma adequação de proteína na dieta Pesquisa de Orçamento Familiar possa promover apropriado crescimento somático da prole.
